The history of the Church of Nuestra Señora de la O is intrinsically linked to the history of Sanlúcar de Barrameda. There is no certainty about its foundation, although tradition and historical studies place it in the year 1360.
It was the noblewoman Isabel de la Cerda y Guzmán, granddaughter of Guzmán el Bueno, who promoted its construction on the site of an old mosque, after the Reconquest of the city.
Over the centuries, the temple has undergone various reforms and expansions that have enriched its architecture and artistic heritage. In 1931, it was declared a National Historical-Artistic Monument, a recognition of its historical and cultural value.

The Church of Nuestra Señora de la O is a building of great architectural interest that combines several styles:
- Mudejar style: The original 14th-century structure preserves Mudejar elements, such as the use of brick and the influence of Islamic architecture.

Beyond its architectural value, the Church of Nuestra Señora de la O is the most important temple of the Holy Week in Sanlúcar de Barrameda.
Declared a Festival of National Tourist Interest of Andalusia in 2002, the Sanlúcar Holy Week has fourteen brotherhoods that make their Penitential Station each year to this parish, the oldest in the city.
Its walls have witnessed key moments of the passion of the brotherhoods, such as the blessing, on 2 March 2003, of the image of the Santísimo Cristo del Perdón, which represents the moment when Jesus is stripped of his garments.
Among the brotherhoods based in this temple, the Royal and Illustrious Brotherhood of the Santísimo Cristo de la Vera Cruz, Nuestra Señora de la Soledad en los Misterios Dolorosos del Santo Rosario and Beato Marcelo Spínola, which processes on Good Friday, stands out.
Its departure takes place at 5:45 PM from Calle Luis de Eguílaz, next to the Church of Nuestra Señora de la O, and returns at midnight, closing the most solemn day of the week.
Also the Very Ancient, Royal and Illustrious Brotherhood and Primitive Archconfraternity of Nazarenes of Our Father Jesus Nazarene, Holy Cross in Jerusalem, Nuestra Señora de la Amargura and Saint John the Evangelist, popularly known as "El Nazareno", has a close link with this temple.
